The ITAM can be part of the same polypeptidic chain that engages the ligand, as in the case of FcγRIIA and FcγRIIC, or of a separate γ subunit that associates noncovalently with the ligand-binding subunit of the receptor, as in the case of FcγRI and FcγRIIIA. Upon receptor clustering, the ITAM associates with and becomes phosphorylated by Hck, Lyn (33, 34), and/or Fgr (35).

In neutrophiles Fgr is physically and functionally associated with Fc gamma RII and involved in Fc gamma RII-mediated signal transduction pathways.
