Protein rpS6/40S subunit map
rpS6 phosphorylation sites have been mapped to five clustered residues that are conserved in metazoans, consisting of Ser235/236/240/244/247, located at the C-terminal part of the protein.

While S6K1 phosphorylates rpS6 at Ser235/236/240/244, RSK exclusively phosphorylates rpS6 at Ser235/236.

The functional role of rpS6 phosphorylation is poorly understood. Recently, it has been revealed that phosphorylation of rpS6 at Ser235/236 promotes its recuitment to the 7-methylguanosine cap complex.
