Cytosolic DNA induces type I IFNs through the endoplasmic reticulum membrane protein STING, which subsequently activates the transcription factors NF-κB and IRF3 through the kinases IKK and TBK1, respectively.
As a high-affinity ligand for STING, 2′3′-cGAMP binds and activates STING through a series of structural changes. Activated STING then recruits TBK1 to phosphorylate IRF3 and activate IKK to phosphorylate IκBα, leading to IκBα degradation.
