TNIP3 (ABIN3‭) ‬inhibits IKK complex trough direct binding to IKBKG.
It results in inhibition of‭ ‬FNKBIA phosphorylation and
proteasomal degradation and prevents activation
of downstream‭ ‬NFkB‭ ‬signaling‭ ‬downstream of IL10.

Agonist stimulation induces IKK2, a catalytic subunit of the IKK complex, to phosphorylate p105. These phosphorylations
create a binding site for the ubiquitin E3 ligase complex SCF-bTrCP which catalyzes K48-linked polyubiquitination of p105, followed by its proteasomal
degradatio. This releases TPL-2 from p105 inhibition and allows TPL-2 to access its substrate MEK
