GM-CSF uniquely inhibited signal transducers and activators of transcription (STAT3) and promoted STAT5 activation, probably downstream of JAK2. STAT5ab(null/null) MDSC were rendered sensitive to sunitinib in the presence of GM-CSF in vitro.

G-CSF downregulates IRF8 expression via STAT3 and GM-CSF via STAT5. IRF8 inhibition provides immunosuppressive functions of MDSC.
